An aster floret cross sections
A cross section through a disc floret, one of 2 dozens or so within a flower bud of some relative of a daisy I found on the campus lawn. Embedded in agarose, sectioned at @200 um, stained with Congo Red and Calcofluor White.

Simply amazing, this is a candidate for Nikon's SmallWorld contest, I think you'll get at least an honourable mention.
This is just beautiful. Did you use a rotary microtome? I would love to learn your staining protocol too.

I'm using Leica Vibratome, it is very versatile, as long as you embed in ~7% agarose and your sections are pretty thick (>50 um). My staining protocol is simple: Calcofluor White (0.1 mg/ml, Sigma-Aldrich #F3543-1G), Congo Red (0.5 mg/mL, Sigma-Aldrich #C676-25G) (I eyeball the amounts at this point ) in PBS-T (PBS + 0.1-0.5% Triton X-100, or any non-ionic detergent). Overnight, RT. Works for cellulose-containing samples as well as for chitin (invertebrate exoskeletons). CR has more apparent affinity for more tightly packed/sclerotized cellulose/chitin. CW stains preferentially (again, my own observation) thinner/more flexible cellulose/chitin. Hope that helps!
