Have you tried the technique of using diffused darkfield illumination, sent in from the sides instead of up through a condenser? --Rik Thanks ... Well I got this far today. I am not sure what the next step is? I didn't want to try any oil objectives until I understand the settings. Getting closer to at least testing the setup. The LED illuminator from Wlodek should be here early next week. The next step will be figuring out the DIC arrangement and how to use it. Second attempt. Image zoomed 100% in PS then cropped, Nikon PlanApo CFN 60x/1.40, oiled slide and condenser, condenser closed to smallest NA, GIF used, then manipulated in PS. The diatom is 55 um in diameter, and the dots were about 0.459 um in diameter. http://www.photomacrography.net/forum/userpix...

I tried to photograph this several times but could never get the sharpness I wanted. This particular try (which was the best) was BF with 'no' GIF. I opened the condenser (from a fully closed position) until what I 'thought' looked the best. Which was very little. Both sides of the slide were oiled ...
