Amber spots?

Found this feller in a sample from a stream that I have had for a little while. I am curious to know what these little dots inside the organism is. I have seen others in the sample, but none with the "spots" on it... 10x 28 image stack http://www.photomacrography.net/forum/userpix/1729_full_10x_1.jp...

Hello Charles! The reason I did this was because the Ikea light was being drowned out by the halogen. When I lowered the halogen it gave out two different lighting situations to deal with. One with the yellowish tint and the more cool led Ikea lamp. I gave up on this terrible illumination technique ...

Hope I didn't squish him too much with the coverslip

Hey folks, I tried out some oblique brightfield lighting...not sure if I pulled it off correctly... I tried using my BH-2 halogen light inside the scope but for some reason I wasn't liking how it was lit. I busted out two Ikea lights and removed my condenser. Placed one in the stead of the condenser...

Sweet Lord Krebs!!!! Those are some nice examples!!!!!! I can't afford DIC (I recently found one for my scope and it mad me feel like I got kicked in the gut) i have seen images like that i had always thought they were DIC and unattainable for me as of now. I was settling for a darkfield condenser.....

- As Charles says you can't do darkfield with a 1.25NA objective, only if it has a built in diaphragm to reduce its NA (but sometimes it may work to get circular oblique illumination) I totally missed that  ....I was reading this on my phone.....Ill use that as my excuse I suppose. ha! It wou...

But I wouldn't go "oiled" unless you really need it for high NA objectives (although in some cases you do need it just for that reason). Would you consider my 100x NA 1.25 high, or were you speaking of something more along the lines of a higher NA? I'm interested in getting the Rotifers and such wh...

Ok, I think I understand. As far as quality goes, is oil better or is a dry darkfield better? Or does it just really come down to preference?

Oil DF or dry DF

As far as I know, this is the place to put this...I have 3 objectives a 10x 40x and a 100x oil. I am going to purchase a Darkfield condenser this weekend, hopefully. Will the oil darkfield produce better results? Can I use the oil darkfield on the non-oil objectives, or do I have to limit the dry da...

If I got that image from 100X I would be lark happy. This is good news!!! Makes me totally happy! I do see some haloing around that small curly fiber that might possibly be reduced with better tuning of the sharpening. Or maybe it's an optical effect. Light does pretty weird stuff down at this scal...

Another annoying stacking question

Bwaa ha ha! It's me again with a 100x image that is a 108 image stack from an oil immersion objective. This is a section of the tarsus on a cockroach that sadly did not make it across the bathroom sink. Poor guy, or gal. I had to sharpen the snot out of this photo and raise the contrast 20% to get i...
