Re: Batch processing multiple stacks located in the same folder using Zerene Stacker

So, I am now considering what a good practical facility should look like, if I were to add one into Zerene Stacker. I would be interested to hear more from you and other people about what data organizations you have and what you'd like to do with them. I really would love to see a batch facility in...

Re: New guy here with Olympus question

Regarding ergonomics: The biggest thing for me in the BX world were the extra thick rubber sleeves for the coaxial table drive.
In recent times there was an add-on to extend the standard height (BX43/53 and surely BX40) table controls down to desk height. This is also very comfortable.

Re: New guy here with Olympus question

Hi, i cant really follow why the BX40 stand is regarded as so much inferior. The light source is lower power (as is the source in the BX41 and 43) and the objective revolver is not interchangable in the default configuration. Also in contrast to the BX50 it does not have filters in the transmitted i...

Re: (Help) DIY motorized microscope stage

Hi,
i cannot give you first hand experience (i use a motorized prior stage) but these might give you some pointers:

Re: Amateur uses for fluorescence microscopy?

You can get pretty far with micro-manager (https://micro-manager.org/) its a free and open source control software for microscopes (everything from hobby usb cameras to fancy custom build setups). chances are good that you can get the fw camera working. it has a broad range of drivers (also industri...

Re: Amateur uses for fluorescence microscopy?

Hi, this looks like a standard setup for in situ hybridisation (e.g. for genetics or pathology). DAPI is used as a counterstain in many fixed pathological/biological samples. Spectrum green and spectrum orange are proprietary fluorophores used by abbott. These filter sets are very "narrow" and give ...

Re: Software question

You need to know how many pixels correspond to a micrometer or vise versa. Best get an objective micrometer, these used to be expensive but now come quite cheaply from china and are adequately for nearly all purposes. Take a picture of the objective micrometer (or any other feature with a known size...

Re: Software question

This might better fit in the technical forum

You need some sort of pixel calibration (know how many µm is your pixel). ImageJ can make this easy for you (https://kaplinskylab.domains.swarthmore ... alebar.htm). If you want to have more aesthetic control you have to do it manually.

Re: Fibroblast cells with and without deconvolution

If someone wants to try deconvolution (and has matlab) at home there is a quite recent publication by Guo et al. "Rapid image deconvolution and multiview fusion for optical microscopy" https://www.nature.com/articles/s41587-020-0560-x which also offers ready to use code. Compared to the previously a...

Re: Olympus U TLU tube lens vs SWTR-2 Superwide Head

Thank you for this test. It is really helpful (i have been one of the guys suggesting that the superwide heads might deliver better quality at the edges of the image but never had the time to formally test it). Officially the TLU is for 22 mm but it odes not surprise me that it works beyond that. ma...

Re: Anyone had any experience with LM Microscope camera adapters?

I have no first hand experience but LM Scope was the company our Olympus technician recommended as first pick for DSLR adaption. Also said that they provide excellent support and reccomendations.
They are pricey though.

If you have Phase contrast a PH3 or PH2 ring should also provide DF on 4x.

Re: fluorescence metoo

I cannot agree more with jmc and others. you should be careful with what you do (and follow the instructions in the manual closely). There are some specifics like "not touching the glass" and not looking into the unshielded lightsource that come with using a arc source. Osram ans Ushio both make bul...

Re: Microscope stage Vs objective Y Axis Movement

The Olympus BX45 and its successor BX46 are modern examples of fixed stage, moving nospiece designs.
There is also an attachmet (WI-NPA) for the BX51/BX61 (and 53, 63) upright that mounts the nosepiece on the stage mount to allow fixed stage designs (used in the water immersion design BX51wi).
