Re: Nikon CFN Fluor / Planapo 100 160/0.17

Is there really such a huge difference in contrast? The specimen in the picture taken with the 40x almost looks translucent compared to the image with the 20x. Most antique slides have coverglasses of about 0,18mm, which is not that cool for e.g a 60 0.85 without correction collar, but my Nikon 60 ...

Re: Nikon CFN Fluor / Planapo 100 160/0.17

Here's some of my observations to share. High NA air objectives are very demanding on the coverslip. If the coverslip is inappropriate, the results will be horrendous. 2.jpg 1.jpg 20x on the top, 40x on the bottom. 20x has an NA of 0.75 while the 40x is at 0.95. Looks like my 40x is broken, it's ord...

Re: First attempt at a thin section with a manual microtome

Look the post "A Rose In Our Garden" (single shots) Thanks again for the insight! I'm not going to deal with weird chemicals with odd symbols as of now, I need a decent space first and some kind of safe to store those nasty looking things. I'm not going to bother with heavy lab microtomes, they are...

Re: First attempt at a thin section with a manual microtome

Hi again, With a manual microtome, the blade and "cutting art" are critical. I never use the blade standard of a microtome. I use a razor blade mounted on a thick handle like glass scrappers to best control Thanks for your info! Very helpful. I watched many videos on microtomes, so I try to level t...

First attempt at a thin section with a manual microtome

Cheap manual slicing microtome came in. Snipped some stems from the garden. Jammed the stem into a carrot and sliced with a razor blade. carrot.jpg Slices aren't thin enough. Claims to do 20um, best I can get is probably 50um. The microtome does feel flimsy, perhaps I can get better with practice an...

Re: Newport Damped Optical Posts

That's one clean and lovely setup! I like it. I agree on the XT66 vs. XT95 point. The XT95 stuff are fully self-contained, and very expensive. Mounting is trivial, burning a hole in the wallet is a matter of just several crucial components. XT66 is far simpler. Additionally to your setup, I would ti...

Video: Exploring a century old I.D. Möller strew slide!

Hi everyone. I present to you a slide that's more than a century old, lots of beautiful diatoms to be seen. This is a freshwater strew mount by one of best mounters/companies back in the days. It's still flawless despite the age! https://youtu.be/6Z2YKFs94ig 4k is still processing at the time of thi...

Re: My setup - need some lighting advice

Off-topic but what's that "skull like" thing in the third photo at the top? It's spooking me out. As for the lighting, it's a good start. The ring light will work on some subjects. You can block out segments of the light so the subject is partially lit with a partial ring, then blend images with var...

Re: What even is this: Plan Apo 40x/1.0 W infinity f=180 unbranded m25 75mm long

"Another water immersion objective with some weird housing: eBay item number:263140217445" Does anyone know what WLSM means on that objective? W means water. LSM means laser scanning microscope. It's an objective optimised for confocal (CLSM, FV1000 unit for Olympus). While not labelled UV, I would...

Re: What even is this: Plan Apo 40x/1.0 W infinity f=180 unbranded m25 75mm long

Glad I decided against bidding, else we'd be in a direct bid war. Check this out: eBay item number:224233184762 An Oly 60x SApo with a slot cut out in the front. I asked the seller about it. Hi, what happened to the front? A slot was machined out… is it to dispense water? I'm baffled. Yes, It was or...

Re: Monochromatic Microscopes?

I've tried some of those objectives, and a friend owns a bunch of them. Some are unwieldy beasts, extremely heavy and mounting is far from trivial. I tried out a 5x Zeiss something, it was huge, threadless, shaped like a fat bowling pin. My friend uses some sort of collar to mount them. Definitely a...
