Re: Some autofluorescence

Thank you! Now I learned that there is a British Pteridological Society - at first glimpse I thought this might be something Monty Python related ;)! The Filter set itself is not really "special", it is basically a UV exitation / long pass filter. Here are the specs: https://www.optolongfilter.com/e...

Re: Some autofluorescence

Thank you! The indusiae of the farn were still completely closed. It was surprising, that the fluorescence of the interior was so clearly visible, refracted by the transparent hull. Edit - Sorry, confused it with the moss! The Moss was very old and dry when I took the pictures, so maybe quite some d...

Some autofluorescence

Hello! Optolong was so kind to send me a sample of their Longpass DAPI Single Band Filter Set for testing purposes, and I am really happy with the results. It yields brighter fluorescence than my U-MWU2 from Olympus, which is similar in specs, but might be a bit aged already as excuse. I went throug...

Hey Lou, here's the full image scaled down: http://www.photomacrography.net/forum/userpix/5697_fluo_120x_06_full_1024_1.jpg and here's a link to the full res: https://imgur.com/a/h9ZZM8V Full res scale is 20nm per pixel. You see some irregularity, but I cannot discern the really cool structure, that...

I did an epi fluorescence image of the iridescent scales. This is done with blue excitation, an PlanApoN 60x/1.42 oil, specimen was mounted in immersion oil. I was trying out different techniques, this returned the highest resolution. http://www.photomacrography.net/forum/userpix/5697_fluo_120x_06_c...

Hi Boron,

this was the version with oiled condenser 1.4 top lens.

To quote one of my favorite movies: "I am an oil man"

Thanks for the replies!

@James
Microscope was a Oly BX52, PlanApo N 60x 1.42.
I used an astronomy mono camera, ZWO ASI 183MM Pro, 2.4µm pixel size.
First four images were taken with DIC HR, last two with circular oblique lighting @ 450nm.

Whohoo a whoppin' 3.2x intermediate... That really cuts down your field of view though - but the detail is awesome!

I'd say: QED ;).
