Have you ever tried different excitation wavelengths with your setup? Do you have to refocus the grating for this? Peter I haven't tried different wavelenghs, but that wouldn't be a problem. Any inchoerent light source is very suitable for structured illumination. Refocusing the grating is very eas...

Amazing shots!

How did you prepare the samples? Are them under a coverslip or dry?

Thanks for your answer Jacek! Those are excellent objectives. I also have one Nikon planapo CFN 20x and it's amazing! I mean the "pads" you are talking about here: "Water mites is a preparation on the pads, so they do not crush, I put two pads of thick cover slips. On top put a thin glass 0.17 mm. "...

Very beautiful shots.

Could you give some more information about your setup?

Could you post a picture of your microscope?

What's the brand/specs of your 6,3x objective?

What are the "pads" you are using to prepare the specimens?

Some arranged Diatoms slides

Hello! I've been practicing this technique in the last months and so far these are some of the slides I menaged to make. Unfortunatly my low magnification setup is not good and pictures' quality is low, but they can still give an idea of the composition! http://www.photomacrography.net/forum/userpix...
