Looking for microscope upgrade

We currently are using a Leica Wild M420 dissecting microscope with trinocular mount and Canon 6D. Ideally I would like to achieve better optical quality with a zoom 3X-100X apochromatic. I know that is asking a lot but am wondering if one exists yet. Since 100% of my imaging is for saving photos I ...

Canon software & microscopes

I'm currently using the Canon 6D on three different microscopes. Ortholux II for brightfield and polarized, Wilde M-5 with trinocular for specimens, and Reichert darkfield. I try to make the original image suitable for peer review publication (they don't like touchups or loss from JPEG) and therefor...

Meta Data and Zerene Stacker

When converting RAW Canon images to 16 bit Tiff using Canon's Digital Photo Professions 4 I need to leave off the meta data so that Zerene stacker can do its stacking. Is there a way to reconnect the meta data to the stacked image?

RAW vs. Tiff

To confirm your math I notice that 20MP RAW files become 59 MP files when converted to 16 bitt TIFF.

calibration?

Thanks for the RAW-TIFF information and also the link to the larger discussion which better explains the inner workings of the issue. Some of the images I generate will be used for peer reviewed botanical journals which generally want no image manipulation other than white balance and maybe gamma ad...

Calibration?

Can anyone suggest some good microscope calibration software that will work well with Canon EOS utility in "Live View"? Also just wondering if Zerene Stacker will ever be able to process RAW images instead of requiring a 16 bit TIFF conversion? It seems like 16 bit Tiff is an older file structure th...

Polarized light for Astrosclereid

My analyzer is an unmarked glass and my polarizer is mounted glass but both are of unknown origin to me.

Polarized light for Astrosclereid

I am trying to get an astrosclereid on a black background using polarized light but am only able to achieve a dark blue ground. Using an Ortholux II 100X and Canon Eos 6D. Any suggestions?

I really appreciate all the discussion that is providing me useful insight. I don't believe I will have a $10,000 budget for purchase and would not plan on putting in 100 hours of set up or adaptation time. We do have a local microscope maintenance and repair company which make me more comfortable i...

Fluorescence imaging

Vishnu

The cooled cameras offered in the astronomy online magazines look interesting. Do you think they will likely work in a fluorescence microscope with regards to tube size and proper filed of view. I would need them to work with alignment software and measuring software as well.

Rik Yes, 300 dpi and HDTV quality is what I would be going for. I will not be in an interactive classroom but will be changing magnification with some frequency. I did read your "low power microscope objectives" posts which were interesting and informative but I probably will be using a microscope o...

Need to purchase DSLR and dissecting microscope

I need publication grade digital images and high resolution class room projection digital images of botanical specimens. I prefer to work in a MAC environment so I'm thinking the Canon software with a Canon DSLR would be suitable but I am open to other options if someone know of better performance. ...

:? I'm curious as to why the two different architectures exist of dedicated microscopic cameras and DSLR's since both employ the ccd/cmos technologies and why the large difference in cost?

Thank you for your reply.

We don't do confocal but I will enquire about the multi staining.
